چکیده

Hrd1p and Doa10p organize factors used for the recognition, ubiquitination, and extraction of substrates. Indeed, Hrd1p provides the direct physical link between recognition events in the ER lumen and ubiquitination and extraction activities in the cytosol (Carvalho et al., 2006; Denic et al., 2006; Gauss et al., 2006). However, many pressing questions remain. Left out of the discussion is a second nonredundant lectin, Htm1p (EDEM in mammals), which is crucial in ERAD-L. Also, how substrates are moved to the cytosol remains elusive. The identity of the dislocation pore in ERAD-L remains controversial and whether such a conduit is even needed in ERAD-C and ERAD-M is unknown. Unlike the Hrd1p complex, there are no clear ideas on how the Doa10p complex participates in selecting substrates. There is no doubt, however, that the crucial framework these new studies provides will accelerate the resolution of these and other mysteries of how molecular inspectors carry out their work in ER quality control.

Interaction of proliferating cell nuclear antigen with PMS2 is required for MutLα activation and function in mismatch repair.

Eukaryotic MutLα (mammalian MLH1-PMS2 heterodimer; MLH1-PMS1 in yeast) functions in early steps of mismatch repair as a latent endonuclease that requires a mismatch, MutSα/β, and DNA-loaded proliferating cell nuclear antigen (PCNA) for activation. We show here that human PCNA and MutLα interact specifically but weakly in solution to form a complex of approximately 1:1 stoichiometry that depends...

Single-molecule imaging reveals target-search mechanisms during DNA mismatch repair.

The ability of proteins to locate specific targets among a vast excess of nonspecific DNA is a fundamental theme in biology. Basic principles governing these search mechanisms remain poorly understood, and no study has provided direct visualization of single proteins searching for and engaging target sites. Here we use the postreplicative mismatch repair proteins MutSα and MutLα as model system...

Target search dynamics during post-replicative mismatch repair

Diffusion is a fundamental physical phenomenon that underlies the molecular basis of all biology. For example, virtually all aspects of DNA metabolism require proteins to search through a vast sea of nonspecific DNA to locate specific binding targets, and these searches are governed by diffusion. An effective search relies on the ability of a protein to efficiently locate and recognize specific...

Mismatch Repair-dependent Iterative Excision at Irreparable O- Methylguanine Lesions in Human Nuclear Extracts*

The response of mammalian cells to SN1 DNA methylators depends on functional MutSα and MutLα . Cells deficient in either of these activities are resistant to the cytotoxic effects of this class of chemotherapeutic drug. Because killing by SN1 methylators has been attributed to O-methylguanine (MeG), we have constructed nicked circular heteroduplexes that contain a single MeG-T mispair and have ...

Schizosaccharomyces pombe MutSα and MutLα Maintain Stability of Tetra-Nucleotide Repeats and Msh3 of Hepta-Nucleotide Repeats

Defective mismatch repair (MMR) in humans is associated with colon cancer and instability of microsatellites, that is, DNA sequences with one or several nucleotides repeated. Key factors of eukaryotic MMR are the heterodimers MutSα (Msh2-Msh6), which recognizes base-base mismatches and unpaired nucleotides in DNA, and MutLα (Mlh1-Pms1), which facilitates downstream steps. In addition, MutSβ (Ms...
